• 1.Mix fresh anticoagulated blood with the erythrocyte sedimentation solution at a ratio of 4:1. Let the mixture stand at room temperature for 30 minutes to allow the sedimentation of red blood cells.2.Transfer a specific volume of the separating medium to a centrifuge tube, carefully lay the supernatant from the sedimented blood onto the surface of the separating medium, and centrifuge at 800 g for 20 minutes to obtain a high-purity layer of individual nucleated cells. The volume ratio of the separating medium to the original whole blood should be 1:1 to 1:2. Refer to the separating medium manual for specific separation experiment steps.3.Wash the obtained high-purity individual nucleated cells twice with PBS or physiological saline.
