Seperation Tube for Human Peripheral Blood
Pour blood, as shown in Figure 1 (B). The blood sample must be anticoagulant blood without the need of dilution. Centrifuge at 800 g for 15 min at 20℃. After suctioning out a portion of the plasma, directly pour the remaining small amount of plasma, PBMC layer, and a small amount of separation medium above the porous barrier into a clean centrifuge tube. As shown in Figure 1 (C), and the PBMC layer is below the plasma and above the separation medium.

Immune Cell Culture Media
Immue cell culture is the key process in basic immunology research, biopharma development, and therapeutic development. However, immune cell culture is much more complicated than cell line culture, especially considering mid/large scale production or maintaining cell function thereafter. BioSci provides a series of products covering major areas of immune cell culture, after investigating these key applications: cancer immune therapy such CarT or NK cell therapy, vaccine development, Treg therapy, immune cell model for cytotoxicity, etc.
BioSci launched L500 serum free medium for human lymphocyte, and NK cell culture medium ten years ago. These immune cell culture media gained good feedback on market from various customers. To catch up with the fast development of immune cell therapy, BioSci launched two upgraded NK cell culture media, NK 2.0 kit for peripheral blood and NK 3.0 kit for cord blood. BioSci also launched new CIK inducing kit, upgraded the L520 serum-free medium for T cell culture, and expanded the affiliated supplements for different culture purpose. Other special medium, such as medium for Treg, is underway in R&D.

NK Cell Culture Kit 2.0 and 3.0
Natural killer cells induction culture kit 2.0 and 3.0 are GMP-grade in vitro induced expansion culture kit, which is suitable for natural killer cell expansion without feeder cells. The kit 2.0 is for peripheral blood, and kit 3.0 is for cord blood. Both kits are with US DMF registration. Either Kit contains necessary component for NK culture, eg. NK kit 3.0 includes NK cell coating solution, NK cell induction solution, NK cell activator, NK cell culture supplement and N300 serum-free medium for NK cells. BioSci also provides recommended optional reagent to form a solution package with NK cell culture, eg. lymphocyte seperation medium/tube, and erythrocyte sedimentation solution.
Both kits gain very good market feedback among users when comparing with other suppliers. For example, start with D0 with 2.5×107 UBMC in T25 and cultured for 16-18 days following NK kit 3.0 protocol, a final system is about 1200 ml, 3-5 billion, (total cell multiplication was 100 to 200 times), positive more than 80%, and cell viability maintained above 85%.

CIK Activated Agent Kit and L520 Serum-free Medium
CIK activated agent kit is formulated according to the type and dose of stimulating factors required by the optimized cell culture on the basis of a large number of groping and experiments on CIK cell and T lymphocyte culture, and is mainly used as an activated agent for induction of human CIK cell culture. It can be used in combination with various serum-free or serum-containing culture media to induce activation of CIK cells.
L520 Serum-free Medium is Biosci’s latest version of optimized medium for T cell culture. After collecting various customer data on the market for over ten years with the previous L500 Serum-free Medium for T cells, this upgrade version is by far among the best media available on market.
Separate PBMCs from human peripheral blood in an aseptic condition. Determine the percentage of CD3+ T cells in the PBMC by flow cytometry or other suitable methods. According to the number of T cells, CD3/CD28 magnetic beads are added for T cell sorting from fresh PBMCs. Seed T cells at a concentration of 0.5×106 cells/mL. Three samples are cultured with L520 serum-free medium supplemented with neither autologous plasma nor BioSci’s cell culture supplemental mix. Cell viability, phenotype, and proliferation rate are compared between two groups. The results are as follows.
After 14 days of culture with L520 medium, the proportion of CD3+cell can reach above 95%, the CD3+ cells proliferation could reach more than 700 times, and the cell viability can be maintained at 70%-90%
