< 1 EU/mL (285±20) mOsmol/kg Formula: ρ = 0.206x + 1.0052 (x is the proportion of USM-A, ρ is the density of the mixed separation medium) According to the formula or Table 1, take a certain proportion of USM-A and USM-B, mix evenly, and obtain a separation medium with a determined density for cell separation. The osmolality of the obtained separation medium ranges from 285 to 305 mOsmol/kg, with a density error range of ±0.0005 g/mL. For example, to prepare 4 mL of separation medium with a density of ρ = 1.0810 g/mL for separating lymphocytes from mouse/rat peripheral blood: x=(ρ-1.0052)/0.206 =(1.0810-1.0052)/0.206=36.8% VUSM-A=4 mL×36.8%=1.472 mL,VUSM-B=4 mL-VUSM-A=(4-1.472) mL=2.528 mL 1.472 mL of USM-A was mixed with 2.528 mL of USM-B to obtain 4 mL of separation medium with a density of 1.0810 g/mL.
